L'invention est relative à un procédé de préparation d'un dérivé de combrétastatine (I) ou (II) : comprenant les étapes suivantes : on fait réagir l'halogénure de triaryl(3,4,5-triméthoxybenzyl)phosphonium P3 (III) dans laquelle Ar désigne un groupe aryle choisi parmi phényle ou thiényle, avec P2 de formule (IV) ou bien P'2 de formule (V) de façon à obtenir respectivement le composé P4 ou bien P'4 : (VI), (VII); puis au cours d'une étape de déprotection en présence d'un acide et/ou d'une base, le composé de formule P4 ou P'4 conduit, après une éventuelle étape de purification, au composé de formule (I) ou (II).

PROCEDE DE PREPARATION D'UN DERIVE DE COMBRETASTATINE
La présente demande est relative à un procédé de préparation d'un dérivé de combrétastatine de formule (I) ou (II) :

Les composés (I) ou (II) appartiennent à la famille des dérivés de combrétastatine ou dérivés du stilbène qui sont des composés anticancéreux. Ils sont décrits dans les demandes EP 0731085, EP 1264821 , EP 1068870 et EP 1407784. La préparation de ces dérivés repose à l'une des étapes sur la formation de la double liaison C=C. A cette étape, il

peut se former deux isomères Z et E mais seul l'isomère Z ( ) présente une activité anticancéreuse réellement efficace. Le procédé de préparation doit donc conduire à un rapport Z/E élevé.
Le Demandeur a mis au point un procédé alternatif de préparation des composés (I) ou (II) qui repose sur l'utilisation des intermédiaires P2 ou P'2 décrits ci-dessous. Ce procédé présente l'avantage de repousser l'étape au cours de laquelle se forme un intermédiaire cytotoxique. Ce procédé alternatif présente donc moins d'étapes comprenant des composés toxiques, ce qui facilite sa gestion d'un point de vue industriel.

Les articles J.Fluor.Chem. 2003, 123, 101-108 et Synlett 2006, 18, 2977 décrivent la préparation de combrétastatines utilisant à l'une des étapes une réaction de Wittig. La réaction de Wittig est aussi envisagée dans le brevet US 7265136 ainsi que dans les demandes internationales WO 03/084919 et WO 2009/118474.

PG1 désigne un groupe protecteur de la fonction alcool, boc, Fmoc et CBZ désignent respectivement les groupes tert-butoxycarbonyl, 9-fluorénylméthoxycarbonyl et benzyloxycarbonyle. Un groupe protecteur est une entité chimique qui est introduite sur une molécule lors d'une étape dite de protection par modification d'un groupe chimique permettant d'améliorer la chimiosélectivité d'une réaction en évitant les réactions secondaires non désirées au niveau dudit groupe chimique et qui est libéré lors d'une étape ultérieure dite de déprotection. PG1 peut être par exemple le THP (tétrahydropyrane), le MEM (méthoxyéthoxyméthyl), le boc, trityle ou le groupe acétyle (Ac).
Le couplage (amidification) est conduit avantageusement en présence d'un activateur d'acide. On désigne par activateur d'acide un composé dont la fonction est de rendre la fonction acide -COOH de Pi ou ΡΊ plus réactive en vue de favoriser la formation d'une liaison amide. Pour plus de détails sur les activateurs d'acide, on pourra se référer à la revue ChemFiles Vol.7, N°2, page 3 éditée par la société Aldrich Chemical ou bien à Tetrahedron report N°672, 2004, 60, 2447-2467, « Récent development of peptide coupling reagents in organic synthesis ». L'EDCI (chlorure de 1-(3-diméthylaminopropyl)-3- éthylcarbodiimide), le DCC (dicyclohexylcarbodiimide), le TOTU (o- [éthoxycarbonyl]cyanométhylèneamino)-N,N,N',N'-tétraméthyluronium tétrafluoroborate), le HBTU (o-benzotriazol-1-yl-N,N,N',N'-tétraméthyluronium hexafluorophosphate), le N,N- carbonyldiimidazole sont des exemples d'activateurs d'acide ou l'anhydride de l'acide propane phosphonique (T3P). En présence de l'activateur acide, il peut se former un intermédiaire, isolable ou non, comprenant une fonction acide activée de la forme -COZ ; par exemple, dans le cas du chlorure de pivaloyle, Z représente -OtBu.
Le couplage peut être conduit dans un solvant tel que par exemple un solvant chloré, par ex. le dichlorométhane (DCM), un éther, par ex. le THF ou un solvant aromatique, par ex. le toluène à une température pouvant être comprise entre 0°C et 20°C. étape (ii) : réaction de Wittig entre P2 ou P'2 et l'halogénure de triaryl(3,4,5- triméthoxybenzyl)phosphonium P3 conduisant respectivement à P4 ou P'4. Dans P3, Ar désigne un groupe aryle choisi parmi phényle ou thiényle, éventuellement substitué par un groupe (CrC4)alkyle ou (CrC4)alcoxy. La réaction de Wittig est conduite dans un solvant en présence d'une base. P3 est obtenu par la réaction de l'halogénure de 3,4,5-triméthoxybenzyle avec la triarylphosphine correspondante PAr3. On utilise de préférence un chlorure ou un bromure. Un exemple de P3 est le chlorure de triphényl(3,4,5-triméthoxybenzyl)phosphonium qui est décrit en p.102 de J. Fluor. Chem. 2003, 123, 101-108 ou bien son équivalent bromure qui est décrit en p.15-16 de WO 02/06279.
Le solvant de cette réaction peut être par exemple le toluène, le THF, le diméthylformamide (DM F), le chloroforme, le DCM, le trifluorotoluène, un mélange de ces solvants ou encore un mélange biphasique aqueux, comme par exemple le mélange chloroforme/eau.
La base que l'on utilise est de préférence une base forte comme par exemple le NaHMDS (bis(triméthylsilyl)amidure de sodium ; CAS [1070-89-9]), le KHMDS (bis(triméthylsilyl)amidure de potassium ; CAS [40949-94-8]), le méthoxyde de sodium, l'amidure de sodium, l'hydroxyde de sodium. La base peut être mise en présence du sel de phosphonium P3, puis l'aldéhyde P2 ou P'2 peut être coulé sur le sel de phosphonium P3, qui aura été mis au préalable en contact avec la base. Selon une variante préférée permettant d'obtenir un rendement plus élevé en P4 ou P'4, on coule la base sur le mélange formé par l'aldéhyde et le sel de phosphonium. La réaction de Wittig peut être conduite à une température généralement comprise entre 0°C et la température de reflux du solvant. étape (iii) : la déprotection de P4 ou P'4 est réalisée en une ou plusieurs étapes et dans des conditions qui dépendent de la nature des groupes protecteurs X et le cas échéant Pd. L'homme du métier peut se référer à « Greene's Protective Groups in Organic Synthesis », 4eme édition, isbn 978-0-471 -69754-1 pour trouver le cas échéant ces conditions.
Ainsi, pour certains groupes protecteurs (par ex. composé P4 avec X=boc), la déprotection peut être conduite en présence d'un acide AH, organique ou minéral. Dans ce cas, la déprotection conduit au composé P5 sous forme de sel. Pour d'autres groupes protecteurs, la déprotection peut être conduite en présence d'une base B, organique ou minérale. Dans ce cas, la déprotection conduit au composé P'5 sous forme de base. La température de la réaction de déprotection est de préférence comprise entre 0°C et 50°C. L'acide peut être un acide fort tel que par exemple l'acide chlorhydrique, ce qui conduit au chlorhydrate. La base peut être par exemple la soude. Il est possible aussi de combiner un traitement acide et un traitement basique, notamment pour P'4 qui comprend deux groupes protecteurs différents X et Pd , étape (iv) : si nécessaire, l'isomère Z est séparé de l'isomère E par toute technique de purification connue en synthèse organique. Il peut s'agir de la purification par recristallisation utilisant comme solvant un mélange contenant un alcool et une cétone ou un ester et plus particulièrement le mélange méthyléthylcétone (MEC)/eau.
L'étape (iii) ou le cas échéant (iv) peut être éventuellement suivie d'une étape supplémentaire consistant à transformer :
- par ajout d'un acide une combrétastatine sous forme de base (par ex. (Il)) en combrétastine sous forme de sel (par ex.(l))
ou bien par ajout d'une base la combrétastatine sous forme de sel (par ex. (I)) en combrétastatine sous forme de base (par ex. (II)). intermédiaires Pi et ΡΊ
Pi est obtenu selon le Schéma 2 par réaction d'une cétone et d'un dérivé de la L-sérine dont la fonction aminé a été protégée par X.

Le dérivé de la L-sérine des Schémas 2 et 2' peut être commercial (par ex. N-boc-L-sérine) ou accessible facilement à l'aide d'au moins une réaction chimique connue de l'homme du métier (semblable par exemple à celle permettant de préparer la N-boc-L-sérine). [Exemples]
Exemple 1 : préparation du chlorhydrate du composé (II)

Le 3-amino-4-méthoxybenzaldéhyde est obtenu par réduction du composé nitro correspondant selon Tetrahedron Letters 1993, 34(46), 7445-1446.

Avant d'être utilisés, le réacteur est affranchi au DCM, séché sous vide et purgé par balayage d'azote pendant 15 à 30 min, l'erlenmeyer est rincé par du DCM stabilisé amylène puis séché sous azote. On charge dans le réacteur 95 mL de DCM et 34,0 g de Boc-L- sérine-acétonide, on refroidit à 4-10°C et on ajoute à l'aide d'une ampoule de coulée en maintenant la température à 4-10°C, 14,3 g de N-méthylmorpholine. On rince l'ampoule par 2,5 mL de DCM. On ajoute à l'aide de l'ampoule de coulée en maintenant la température à 4-10°C, 17,1 g de chlorure de pivaloyle, on rince l'ampoule par 2,5 mL de DCM. On maintient sous agitation à 4-10°C pendant 2 h.
On prépare sous agitation une solution d'aminobal (20,0 g) dans le DCM (95 mL) et on coule cette solution dans le réacteur en maintenant la température à 4-10°C. On chauffe ensuite le mélange à 20°C en 1 h et maintient sous agitation à 20 °C pendant 16 h minimum. On ajoute dans le réacteur à 20-25°C, 100 mL d'eau déminéralisé, on laisse agiter pendant 20 min et on décante. On soutire la phase inférieure organique, contenant le produit) ainsi que la phase supérieure (majoritairement aqueuse). La phase organique, contenant le produit est rechargée dans le réacteur. On ajoute 140 mL d'une solution aqueuse d'hydroxyde de sodium à 1 ,0 N. On maintient sous agitation à 20-25°C pendant environ 20 min puis on laisser décanter. On soutire la phase inférieure organique, contenant le produit. La phase organique, contenant le produit est rechargée dans le réacteur. On ajoute 100 mL d'eau déminéralisé. On maintient sous agitation à 20-25°C pendant environ 20 min puis on laisser décanter. On soutire la phase inférieure organique, contenant le produit. La phase organique, contenant le produit est rechargée dans le réacteur. On ajoute 100 mL d'isopropanol.
On distiller (35± 5°C dans la double enveloppe) sous une pression résiduelle d'environ 30 mbar jusqu'à avoir un volume résiduel de 100 mL dans le réacteur. On ajuste à 20°C et on laisse agiter à 20°C pendant 3 h. On rince le réacteur et on lave en 2 fois le gâteau avec un volume total de 40 mL d'isopropanol. On sèche le produit à 40°C sous vide 30 mbar. Rendement en produit isolé : 60%. Réaction de Wittig (étape (ii))
On charge dans un réacteur de 7 L : 581 g de sel de phosphonium (1 ,2 éq), 350 g de l'aldéhyde de l'étape précédente (1 ,0 éq) et 3500 mL de CHCI3 (solution jaune-brun soutenu). On ajoute 1 1 10 mL d'une solution NaOH 1 N (1 ,2 éq). On agite vigoureusement le mélange biphasique et la solution devient jaune pâle. On maintient vers 20°C. On ajoute 3500 mL d'eau, on agite et on décante (pH de la phase aqueuse 13). On effectue un 2ème lavage avec 3500 mL d'eau ; le pH est alors de 7. On décante et on soutire la phase organique jaune-orange (volume 4250 mL contenant 346,0 g de Z et 136,7 g de E). Le rapport Z/E est de 72/28 et le rendement en Z+E par rapport à l'aldéhyde est de 96,2%.
La solution est réintroduite dans le réacteur, puis on distille CHCI3 sous vide avec un vide initial de 100 mbar et final de 45 mbar (température double enveloppe vers 30°C). Le mélange devient sirupeux. On casse le vide et on rajoute 50 mL de CHCI3 et 2500 mL d'AcOiPr : on obtient une solution fluide (5250 mL). On reprend la distillation à volume constant avec ajout d'AcOiPr. Il se forme des cristaux (majoritairement de l'oxyde de triphénylphosphine) que l'on filtre. On conserve le filtrat contenant le produit attendu pour l'engager dans l'étape suivante. Rapport Z/E = 71/29. Rendement en Z : 68,9%.
Déprotection en milieu acide (étape (iii))
On charge la solution de l'étape précédente (3045,9 g de solution, soit 343,9 g de Z et 136,9 g de E). On ajoute 295,2 mL d'une solution HCI 12 N (4 éq par rapport au produit). Le mélange biphasique passe du jaune au rouge foncé. On ajoute 1800 mL d'eau, on agite pendant 10 min, on décante et on soutire la phase aqueuse riche. On ajoute 900 mL d'eau à la phase organique. On décante et on soutire la phase aqueuse. On obtient 3714 g de phase aqueuse orange (ratio Z/E = 67/33). On ajoute 2700 mL d'AcOiPr et on coule lentement une solution NaOH 10 N jusqu'à obtenir un pH de 10-1 1. On décante et on soutire la phase aqueuse. On ajoute 2700 mL d'eau et 1 1 g de NaCI et on agite fortement puis on décante. On refait cette opération de battage avec 2700 mL d'eau. On récupère une phase organique jaune (2760 g) ratio Z/E = 68/32. Rendement : 35%.
Recristallisation (étape (iv))
On charge dans un tricol de 250 mL : 5,27 g du produit précédent, 50 mL d'eau, 50 mL d'AcOiPr et 1 ,32 mL de soude à 30%. On agite pendant 30 min. On décante et on soutire la phase aqueuse (pH=10), on effectue deux battages avec de l'eau (50 mL). Après le 2eme battage, le pH est de 7. On évapore à sec la phase organique (40°C, vide de 60 mbar) et on sèche à l'étuve (40°C). On reprend le solide (5,49 g) avec 1 1 ,2 mL de MEC, et on ajoute à la solution 1 ,00 mL d'une solution HCI 12 N (densité=1 ,18). On laisse cristalliser lentement un peu de produit. On rajoute 0,36 mL d'eau et une grande partie du produit cristallisé se redissout. On ajoute alors 2,70 mL de MEC et on laisse cristalliser à nouveau. On agite à l'ambiante pendant 5 jours. On obtient le produit avec un rapport Z/E=93/07. Rendement en Z : 45%. Exemple 1 bis : préparation du chlorhydrate du composé (II) Réaction de Wittig (étape (ii))
On charge dans un réacteur de 500ml_ : 44,8 g de sel de phosphonium (1 ,2 éq), 27 g de l'aldéhyde de l'étape précédente (1 ,0 éq) et 270 mL de CHCI3 (solution jaune-brun soutenu). On ajoute 85,6 mL d'une solution NaOH 1 N (1 ,2 éq). On agite vigoureusement le mélange biphasique et la solution devient jaune pâle. On maintient vers 20°C pendant environ 4h. On ajoute 270 mL d'eau, on agite et on décante (pH de la phase aqueuse 13). On effectue un 2ème lavage avec 270 mL d'eau ; le pH est alors de 7. On décante et on soutire la phase organique jaune-orange (masse 470,4g contenant 26,7 g de Z et 1 1 ,2 g de E). Le rapport Z/E est de 70/30, le rendement en Z+E par rapport à l'aldéhyde est de 98% et le rendement en Z par rapport à l'aldéhyde de 69,0%.
La solution est réintroduite dans le réacteur, puis on effectue un changement de solvant vers l'acétate d'isopropyle sous pression réduite (45 à 100 mbar à 30°C environ). En fin d'opération, on ajuste le volume résiduel à 203mL. Il se forme des cristaux que l'on filtre et lave à l'acétate d'isopropyle. Le filtrat, contenant le produit de réaction est engagé tel quel dans l'étape suivante. Rapport Z/E = 70/30. Rendement en Z : 69,0%.
Déprotection en milieu acide (étape (iii))
On charge la solution de l'étape précédente (248,0 g de solution, soit 26,7 g de Z et 1 1 ,2 g de E) dans un réacteur de 500 mL. On ajoute 23,3 mL d'une solution HCI 12 N (4 éq par rapport au produit). Le mélange biphasique passe du jaune au rouge foncé. On maintien sous agitation à 20°C pendant environ 5 h. On ajoute 137 mL d'eau, on agite pendant 10 min, on décante et on soutire la phase aqueuse riche. On ajoute 69 mL d'eau à la phase organique. On décante et on soutire la phase aqueuse. On obtient 283,6 g de phase aqueuse orange (ratio Z/E = 66/34). On ajoute 206 mL d'AcOiPr et on coule lentement une solution NaOH 10 N jusqu'à obtenir un pH de 10-1 1 . On décante et on soutire la phase aqueuse. On ajoute 206 mL d'eau et 2,1 g de NaCI et on agite fortement puis on décante. On refait cette opération une seconde fois. On récupère une phase organique jaune qu'on amène à sec (35,0 g, ratio Z/E= 66/34). On reprend ce résidu par 108,3 g de MEC. On obtient une solution. On ajoute successivement 5,82 mL d'HCI 12N et 2,75 mL d'eau. On amorce ensuite par addition de 75 mg de l'isomère Z pur. On maintient sous agitation à 20°C pendant 24 h puis on filtre la bouillie obtenue. Le gâteau est essoré à fond puis séché en étuve (50°C, 60 mbar). On obtient ainsi 7, 15g d'une poudre fine beige : rendement en Z : 31 ,5%, ration Z/E=95,9/4,1.
Recristallisation (étape (iv))
On charge dans un ballon de 5 mL : 488 mg de composé I (Z/E=93,5/6,5), 0,1 15 mL d'eau, 268 mL d'acétonitrile. On chauffe à 35°C, on agite jusqu'à obtention d'une solution puis on refroidit à 20°C. A cette température, on amorce par 3 mg de l'isomère Z pur. On maintient sous agitation pendant 30 min puis on coule en environ 2 h 3,44 mL d'acétonitrile. On maintient ensuite sous agitation à 20°C pendant 18 h et on filtre. Le gâteau obtenu est séché à l'étuve (50°C, 60 mbar). On obtient ainsi 367 mg du produit attendu avec un ratio Z/E de 99,65/0,35 soit un rendement de 80%.

4. Procédé selon la revendication 1 à 3 dans lequel PG1 représente l'un des groupes protecteurs suivants : THP (tétrahydropyrane), le MEM (méthoxyéthoxyméthyl), boc, trityle ou acétyle (Ac).
5. Procédé selon la revendication 1 à 4 dans lequel Ar représente le groupe phényle ou thiényle, éventuellement substitué par un groupe (CrC4)alkyle ou (CrC4)alcoxy.
6. Procédé selon l'une des revendications 1 à 5 dans lequel A" désigne CI".
7. Composé de formule P2 :
