Description

PROCEDE DE PREPARATION D'UN DERIVE DE COMBRETASTATINE  PROCESS FOR PREPARING A COMBRETASTATIN DERIVATIVE
La présente demande est relative à un procédé de préparation d'un dérivé de combrétastatine de formule (I) ou (II) : The present application relates to a process for preparing a combretastatin derivative of formula (I) or (II):
A" désignant l'anion associé à un acide AH. Plus particulièrement, A désigne CI". [Problème technique] A " designating the anion associated with an acid AH, more particularly, A designates CI " . [Technical problem]
A cette étape, il Compounds (I) or (II) belong to the family of combretastatin derivatives or stilbene derivatives which are anticancer compounds. They are described in applications EP 0731085, EP 1264821, EP 1068870 and EP 1407784. The preparation of these derivatives is based on one of the steps on the formation of the C = C double bond. At this stage, he
Two isomers Z and E can be formed, but only the Z (isomer) has a truly effective anticancer activity. The preparation process must therefore lead to a high Z / E ratio.
The Applicant has developed an alternative method of preparing compounds (I) or (II) which is based on the use of the intermediates P 2 or P ' 2 described below. This method has the advantage of postponing the step during which a cytotoxic intermediate is formed. This alternative method therefore has fewer steps including toxic compounds, which facilitates its management from an industrial point of view.

Coupling (amidification) is advantageously carried out in the presence of an acid activator. The term "acid activator" denotes a compound whose function is to make the -COOH acid function of Pi or ΡΊ more reactive in order to promote the formation of an amide bond. For more details on the acid activators, reference may be made to ChemFiles Vol.7, No. 2, page 3 edited by Aldrich Chemical or Tetrahedron report No. 672, 2004, 60, 2447- 2467, "Recent Development of Peptide Coupling Reagents in Organic Synthesis". EDCI (1- (3-dimethylaminopropyl) -3-ethylcarbodiimide chloride), DCC (dicyclohexylcarbodiimide), TOTU (o- [ethoxycarbonyl] cyanomethyleneamino) -N, N, N ', N'-tetramethyluronium tetrafluoroborate), HBTU (o-benzotriazol-1-yl-N, N, N ', N'-tetramethyluronium hexafluorophosphate), N, N- carbonyldiimidazole are examples of acid activators or the anhydride of propane phosphonic acid (T3P). In the presence of the acidic activator, an intermediate, isolable or otherwise, may be formed comprising an activated acid function of the form -COZ; for example, in the case of pivaloyl chloride, Z represents -OtBu.
Chem. 2003, 123, 101-108 ou bien son équivalent bromure qui est décrit en p.15-16 de WO 02/06279. The coupling may be conducted in a solvent such as for example a chlorinated solvent, e.g. dichloromethane (DCM), an ether, e.g. THF or an aromatic solvent, e.g. toluene at a temperature of between 0 ° C and 20 ° C. step (ii): Wittig reaction between P 2 or P'2 and the triaryl halide (3,4,5-trimethoxybenzyl) phosphonium P 3 leading respectively to P 4 or P ' 4 . In P3, Ar denotes an aryl group selected from phenyl or thienyl, optionally substituted by a (CrC 4 ) alkyl or (CrC 4 ) alkoxy group. The Wittig reaction is conducted in a solvent in the presence of a base. P 3 is obtained by the reaction of the 3,4,5-trimethoxybenzyl halide with the corresponding triarylphosphine PAr 3 . A chloride or bromide is preferably used. An example of P 3 is triphenyl (3,4,5-trimethoxybenzyl) phosphonium chloride which is described in p.102 of J. Fluor. Chem. 2003, 123, 101-108 or its bromide equivalent which is described in p.15-16 of WO 02/06279.
The solvent of this reaction may be, for example, toluene, THF, dimethylformamide (DM F), chloroform, DCM, trifluorotoluene, a mixture of these solvents or an aqueous biphasic mixture, for example the chloroform / water.
The base used is preferably a strong base such as, for example, NaHMDS (sodium bis (trimethylsilyl) amide CAS [1070-89-9]), KHMDS (potassium bis (trimethylsilyl) amide, CAS [ 40949-94-8]), sodium methoxide, sodium amide, sodium hydroxide. The base may be brought into contact with the phosphonium salt P 3 , and then the aldehyde P 2 or P ' 2 may be cast on the phosphonium salt P 3 , which has been brought into contact with the base beforehand. According to a preferred variant making it possible to obtain a higher yield of P 4 or P ' 4 , the base is cast on the mixture formed by the aldehyde and the phosphonium salt. The Wittig reaction can be conducted at a temperature generally between 0 ° C and the reflux temperature of the solvent. step (iii): the deprotection of P 4 or P ' 4 is carried out in one or more steps and under conditions which depend on the nature of the protecting groups X and, where appropriate, Pd. The skilled person may refer to "Greene's Protective Groups in Organic Synthesis", 4th edition, isbn 978-0-471 -69,754 to 1 to find if any of these conditions.
Thus, for certain protecting groups (eg compound P 4 with X = boc), the deprotection can be conducted in the presence of an organic or inorganic HA acid. In this case, the deprotection leads to the compound P 5 in salt form. For other protecting groups, the deprotection can be conducted in the presence of a base B, organic or mineral. In this case, the deprotection leads to the compound P ' 5 in base form. The temperature of the deprotection reaction is preferably from 0 ° C to 50 ° C. The acid may be a strong acid such as for example hydrochloric acid, which leads to the hydrochloride. The base may be, for example, sodium hydroxide. It is also possible to combine an acid treatment and a basic treatment, especially for P ' 4 which comprises two different protective groups X and Pd, step (iv): if necessary, the Z isomer is separated from the E isomer by any purification technique known in organic synthesis. This may be the purification by recrystallization using as solvent a mixture containing an alcohol and a ketone or an ester and more particularly the methyl ethyl ketone (MEK) / water mixture.
L'étape (iii) ou le cas échéant (iv) peut être éventuellement suivie d'une étape supplémentaire consistant à transformer : Step (iii) or where appropriate (iv) may possibly be followed by an additional step of transforming:
(Il)) en combrétastine sous forme de sel (par ex.(l)) - by addition of an acid combretastatin in base form (eg (II)) to combretastine in the form of salt (eg (1))
(II)). intermédiaires Pi et ΡΊ or by addition of a base combretastatin salt (eg (I)) combretastatin base (eg (II)). Intermediate Pi and ΡΊ

On obtient ainsi 7, 15g d'une poudre fine beige : rendement en Z : 31 ,5%, ration Z/E=95,9/4,1. The solution of the preceding step (248.0 g of solution, ie 26.7 g of Z and 11.2 g of E) is charged to a 500 ml reactor. 23.3 ml of a 12 N HCl solution (4 eq with respect to the product) are added. The biphasic mixture changes from yellow to dark red. Stirring is continued at 20 ° C. for approximately 5 hours. 137 ml of water are added, the mixture is stirred for 10 minutes, the mixture is decanted and the rich aqueous phase is withdrawn. 69 ml of water are added to the organic phase. The mixture is decanted and the aqueous phase is withdrawn. 283.6 g of orange aqueous phase are obtained (Z / E ratio = 66/34). 206 ml of AcOiPr are added and a 10 N NaOH solution is slowly poured until a pH of 10-1 1 is obtained. The mixture is decanted and the aqueous phase is withdrawn. 206 ml of water and 2.1 g of NaCl are added and the mixture is stirred vigorously and decanted. We repeat this operation a second time. A yellow organic phase is recovered which is brought to dryness (35.0 g, ratio Z / E = 66/34). This residue is taken up with 108.3 g of MEK. A solution is obtained. 5.82 ml of 12N HCl and 2.75 ml of water are successively added. It is then initiated by adding 75 mg of the pure Z isomer. Stirring is maintained at 20 ° C for 24 h and then the slurry obtained is filtered. The cake is wrung thoroughly and then dried in an oven (50 ° C, 60 mbar). There is thus obtained 15 g of a fine beige powder: yield Z: 31.5%, ration Z / E = 95.9 / 4.1.
Charge in a 5 ml flask: 488 mg of compound I (Z / E = 93.5 / 6.5), 0.115 mL of water, 268 mL of acetonitrile. The mixture is heated to 35 ° C., stirred until a solution is obtained and then cooled to 20 ° C. At this temperature, it is initiated with 3 mg of the pure Z isomer. Stirring is maintained for 30 min and then poured in about 2 hours 3.44 mL of acetonitrile. It is then stirred at 20 ° C. for 18 h and filtered. The cake obtained is dried in an oven (50 ° C, 60 mbar). 367 mg of the expected product is thus obtained with a Z / E ratio of 99.65 / 0.35, ie a yield of 80%.
